CARsgen Presents First-in-human Results of GPRC5D Targeting CAR-T CT071 at EHA 2024

SHANGHAI, June 15, 2024 /PRNewswire/ — CARsgen Therapeutics Holdings Limited (Stock Code: 2171.HK), a company focused on innovative CAR T-cell therapies for the treatment of hematologic malignancies and solid tumors, announces that the initial results from the ongoing first-in-human study of CT071 have been presented at the 29th Annual Congress of the European Hematology Association (EHA).

The preliminary results of Phase I of CT071 (NCT05838131) were presented as a poster at the 29th EHA Annual Congress on June 14, 18:00 – 19:00 CEST, which was titled “First-in-human study of GPRC5D-targeted CAR T cells (CT071) with an accelerated manufacturing process in patients with relapsed/ refractory multiple myeloma (RRMM)”.[1]

“Multiple myeloma is a common yet incurable hematologic malignancy with high unmet need. Despite the numerous recent advances, most patients relapse and become refractory to available therapies and therefore, novel therapies are needed. GPRC5D, a protein highly expressed on the surface of malignant plasma cells with limited expression on normal tissues, represents a promising target for treating multiple myeloma. CT071 is a fully human GPRC5D-targeting autologous second-generation CAR T-cell product manufactured using our expedited CARcelerateTM platform that shortens the manufacturing time to around 30 hours, supporting a shorter vein-to-vein time and younger T cells. The preliminary results of the ongoing study presented at EHA shows that CT071 has the potential to be the best-in-class GPRC5D targeting CAR-T therapy,” said Raffaele Baffa, M.D., Ph.D., Chief Medical Officer of CARsgen Therapeutics. “We are excited about advancing CT071 and look forward to sharing future updates with the medical community.”

As of February 28, 2024, 10 patients were dosed with CT071—7 patients at 1.0×105 cells/kg and 3 patients at 3.0×105 cells/kg. Among them, 80% had high-risk cytogenetics, 30% had one or more extramedullary plasmacytomas (EMD), and 40% were at R-ISS stage III. This was a heavily pre-treated population with a median of 5 prior lines of therapy, including 90% double-class refractory, 70% triple-class refractory, 40% penta-drug refractory, 50% having received autologous stem cell transplantation, and 20% had previously been treated with BCMA/CD19 dual-targeting CAR T cells. None of the patients on the study required bridging therapy due to rapid manufacturing turnaround.

The median follow-up at the time of data cut-off was 4.07 months (range: 2.8-7.4). There were no Grade 3 or higher cytokine release syndrome (CRS) events. No immune effector cell-associated neurotoxicity syndrome (ICANS) was observed. No adverse events of special interest or dose limiting toxicity (DLT) occurred. Four patients experienced treatment-related SAE, including pneumonia (n=1), decreased appetite (n=1) and thrombocytopenia (n=2), and all recovered.

The overall response rate was 90%, including 5 patients (50%) with stringent complete response (sCR), 2 patients (20%) with very good partial response (VGPR), and 2 patients (20%) with partial response (PR). All the 9 patients with evaluable MRD assessment at Week 4 achieved MRD negativity (10-6 threshold) ), including all 5 patients with sCR/CR. The pharmacokinetic analysis demonstrated robust cell expansion and persistence, with median Tmax of 14 days (range: 12-28) and median Cmax of 32280.5 copies/μg gDNA (range: 8372-106060).

CT071 is a CAR T-cell therapy candidate developed utilizing CARsgen’s proprietary CARcelerateTM platform targeting GPRC5D for the treatment of R/R MM or relapsed/refractory plasma cell leukemia (“R/R PCL”). An investigator-initiated trial (NCT05838131) is ongoing in China to evaluate the safety and efficacy of CT071 for the treatment of R/R MM or R/R PCL. 1. What is San Miguel Chapel famous for? 

The San Miguel Chapel is famous for being known as the oldest church in the United States. 

The church is located in Santa Fe, New Mexico. 

It was constructed “under the direction of Franciscan friars to serve a small congregation of soldiers, laborers, and Indians who lived in the Barrio de Analco,” per the church’s website.

The first historical written record of the church dates back to 1628.

Yet many believe that the construction of the church dates back even further, according to the San Miguel Chapel website, with oral history putting the church’s creation around 1610, in tandem with the founding of Santa Fe.

The chapel was dedicated to Archangel Michael. 

A statue of him stands at the site of the church today, according to U.S. News & World Report. 

2. What happened to San Miguel Chapel? 

San Miguel Chapel has undergone reconstruction over the years. 

Documentation retained by the church, according to its website, shows that the chapel was heavily damaged, including the roof being burned during the Pueblo Revolt of 1680, also known as Popé’s Rebellion. 

Visitors can view the 780 pound San Jose Bell that used to be in the bell tower. 

In 1710, San Miguel Chapel was rebuilt. Further repairs funded by the mayor of Santa Fe were done in 1798, according to the church’s website. 

The community came together in 1887 for another rebuild when the church was in need of so many repairs, it was nearly demolished.

While the rebuilding of San Miguel Chapel has changed the structure from what it was when it was originally built, the church is still full of history. 

For example, parts of the original walls still remain intact.

Additionally, guests can view the 780 pound San Jose Bell that used to be in the bell tower. 

Sadly, the tower fell in a storm back in 1872 — but the bell is still on display inside the chapel.

China’s Hainan FTP Cements ties with South Korea and the United States

British couple traveling to Spain mistakenly flown to Lithuania after being escorted to wrong plane

A British couple planning to travel to Spain for a family vacation was instead flown 1,500 miles away to Lithuania after airport staff escorted them to the wrong aircraft.

Andrew and Victoria Gore, both 47, intended to board a flight to Barcelona and then head to Costa Brava on May 25 to celebrate Mr. Gore’s 47th birthday. But they boarded the wrong flight with the budget airline Ryanair, as the rest of their family made it to Barcelona, according to BBC News.

The couple told BBC News they were feeling “distraught” after the mixup, which they described as a “nightmare.”

“We were told they were definitely on the flight and they said ‘just relax’, so we had a sleep as we’d been up early,” Mr. Gore told the outlet. “When I woke up, it didn’t look like Spain out the window, I turned my phone on and it said ‘Welcome to Lithuania.'”

Mr. Gore, an amputee, and Mrs. Gore, who is autistic, ended up on a flight to Kaunas, Lithuania, after booking special assistance at Bristol Airport in England. Passengers with disabilities or reduced mobility are legally entitled to special assistance at airports without additional charge when traveling from various regions, including the U.K. and the U.S.

“We have been away lots of times and always had special assistance, so this was nothing new to us,” Mrs. Gore told BBC News. “The minibus took us to the Ryanair plane and they checked our boarding pass and let us on the plane.”

Bristol Airport said it was working to investigate what happened and to make improvements to ensure this type of incident does not happen again, while Ryanair said crews make several announcements before departure about the flight destination and attributed the error to Bristol Airport’s special assistance provider, BBC News reported.

A spokesperson for Ryanair said that Bristol Airport’s special assistance provider boarded the couple onto the wrong flight to Kaunas “despite gate signage clearly displaying the flight’s destination.” 

“Before departing, crew make several announcements advising passengers of the flight’s destination,” the spokesperson said, adding that they “sincerely apologize to these passengers for any inconvenience.”

A Bristol Airport spokesperson said: “We’ve worked with our business partners to investigate this incident. All customers have their travel documentation checked by the airline or their ground handling agent before boarding an aircraft.”

“Since being informed of the issue, Bristol Airport has worked with our airline handling agent and special assistance provider to investigate the circumstances and to introduce improvements for the future,” the spokesperson added. “We will contact the customer with information to direct their complaint to the correct business partner for resolution.”

“Our flight number came up at the gate, and we were taken to the bottom of the plane steps, where the ground staff checked our boarding passes,” Mr. Gore said. “When we got on the plane, the stewardess checked our passes again.”

After alerting Ryanair officials that they were in the wrong country, they were booked on the next available flight to Barcelona, which was not until the next morning and from Riga in the neighboring country of Latvia.

The airline booked the couple overnight accommodation and a 150-mile Uber ride to the airport.

They eventually landed in Barcelona and made it to their hotel on the Costa Brava, but their bags did not make it until two days later, the outlet noted, since they did not board their initial flight from Bristol, England.

“It was surreal, and an experience I never want to have ever again. Our family were worried sick, and we were exhausted when we did eventually arrive,” Mr. Gore said. “Even though they took us to the right place eventually, the experience had already happened, the damage had been done, and we can’t forget those worrying moments.”

Taking a vacation from work may soon become mandatory

With burnout prevalent in the workplace, some employers are considering mandatory vacation, full office shutdowns and other ways to get workaholic employees to take a break.

Only a small number of employers today — 8.7% — require workers to take a minimum amount of vacation, and some of this is for legal or regulatory reasons, according to a 2024 report by the International Foundation of Employee Benefit Plans. Even so, figuring out how to get employees to take time off is increasingly on employers’ minds, said Julie Stich, vice president of content at the International Foundation of Employee Benefit Plans. 

And for good reason. A recent Harris Poll survey found that 78% of Americans do not use the maximum amount of paid time off allowed by their employer. The average worker took 15 paid days off last year, despite nearly half being allowed more than that by their employer, the poll found. 

Yet burnout rates are high. Forty-five percent of American employees claim to feel burnout at work, according to a 2023 survey by Eagle Hill Consulting. That’s even higher among 18 to 32 year-old workers (52%) and women (48%).

“There’s more discussion about mandatory time off than there was in the past. There are a lot of instances when companies are reporting employees are not taking time off,” Stich said.

Here’s what companies and employees grappling with these issues need to think about.

Tracking vacation usage and employee mindset

An important first step is to understand the company’s baseline, said Wende Smith, head of people operations at BambooHR. This can help companies pinpoint a plan for moving forward. Is the problem only in a particular department or is it more broad-based?

“If you’re not tracking it, you won’t understand what your problem is, or if you have a problem,” Smith said. 

Before enacting broad policy changes, it’s important to address the underlying issues that might be preventing employees from taking time off to recharge.

This could include heavy workload, lack of adequate staffing and the need to coordinate time off with other staff members, according to the International Foundation of Employee Benefit Plans’ survey.

If employers are really serious about getting employers to take time off, they need to talk to employees, find out their reasons they aren’t taking vacation, and address them, Smith said. Leaders also need to be a good role model for their direct reports in terms of vacation-taking, she said.

And, when employees do take vacation, respect their time off, Stich said. “Don’t constantly message them while they are on vacation.”

Mandatory paid leave pros and cons

Jobs such as pilots, air traffic controllers, trucking and certain finance roles may have mandatory time off by law or by industry best practice, Stich said. But other industries may have more flexibility when it comes to setting these policies. Mandatory policies can be beneficial because they force the issue of vacation, but there can be challenges as well, such as employee pushback and enforcement, said Jonathan Gove, senior executive advisor at Eagle Hill Consulting.

“There might be some people who see that vacation time as a bit of a savings account, especially if they get paid out at termination,” said Kate Derby, West region practice lead for the absence, disability management and life specialty practice at WTW.

Additionally, mandatory time-off policies can be complicated to administer. If an employer is going to put this into place, they have to define how many days must be taken off, Stich said. Is it one day off a month, one day per quarter, or five days off in a row? Use of blackout periods also must be weighed for a business to function properly — for instance, during tax season for accountants, or three weeks before school at a pediatric office when back-to-school checkup season requires an all-hands-on-deck approach.

To avoid scheduling headaches, companies might insist employees tell their manager what week they plan to take off by a certain date, and if they don’t, the manager needs to work with the employee to determine the time off, Stich said.

Companies also have to be prepared to administer, in a way that’s fair, who gets to take the prime weeks because otherwise you could run into legal troubles or have difficulty with disgruntled and disengaged workers, said Mary Will, deputy general counsel and partner at Faegre Drinker. Employers have to be prepared to manage complaints that “Joe always gets to take the week between Christmas and New Year’s and no one else can and we don’t think that’s fair,” she said.

Federal, state and local law

There is no federal law that says employers need to provide paid time-off, but there are some state and local laws that apply. Different states also have different requirements on paying for unused paid-time off, which can be complicated to address with a mandatory vacation policy, said Jill Kahn Marshall, an employment and labor attorney and partner at Reavis Page Jump. Before implementing a new policy, businesses should check with an attorney to make sure they aren’t accidentally running afoul of any state and local laws, she said.

Methods to encourage more time off from work

Instead of focusing on mandatory time off, some companies are looking for less administratively burdensome and employee-friendly alternatives. Many high-tech companies in California, for example, have adopted shut-downs, which typically take place the week of July 4th or between Christmas and New Year’s, said WTW’s Derby. “Even the tech companies that have unlimited PTO are doing shut-downs. It’s the chance to catch-up and catch your breath because everybody is taking the same week off,” Derby said. 

More companies are also scrapping their unlimited time-off policies, which haven’t always worked out as anticipated. In some cases, employees have abused the policy, but often, they take way less vacation than they did when they had a set number of days. As a result, some employers have gone back or are considering returning to policies where employees only accrue a certain number of days — often two to three weeks per year, Will said.

Sander VanderWerf, senior vice president of Aon’s time away and life solutions practice, says some employers may want to consider a vacation plan that requires workers to use it or lose the days. “Employees are more likely to use more time when the time does not carry over at year-end,” she wrote in an email.

ROCKVILLE, Md. and SUZHOU, China, June 14, 2024 /PRNewswire/ — Ascentage Pharma (6855 HK), a global biopharmaceutical company engaged in developing novel therapies for cancer, chronic hepatitis B (CHB), and age-related diseases, announced today the signing of an option agreement with Takeda to enter into an exclusive license agreement for olverembatinib, an oral, potentially best-in-class third-generation BCR-ABL tyrosine kinase inhibitor (TKI), which is currently in development for chronic myeloid leukemia (CML) and other hematological cancers. If exercised, the option would allow Takeda to license exclusive global rights to develop and commercialize olverembatinib in all territories outside of among others, mainland China, Hong Kong, Macau, Taiwan, China.

Despite the impact TKIs have had in the treatment of patients with CML, there remains a significant unmet need for patients whose disease is resistant to these therapies or who develop hard-to-treat mutations following these treatments.

Under the terms of this option agreement, Ascentage Pharma is set to receive 100 million USD from Takeda to acquire the option to license olverembatinib. Ascentage will also be eligible for an option exercise fee and additional potential milestones of up to approximately 1.2 billion USD. Additionally, Ascentage Pharma is expected to receive a minority equity investment from Takeda.

As part of the agreement, Ascentage Pharma will continue to be solely responsible for all clinical development of olverembatinib before the potential exercise of the option to license olverembatinib. Olverembatinib is currently approved and marketed in China for the treatment of adult patients with TKI-resistant chronic-phase CML (CP-CML) or accelerated-phase CML (AP-CML) harboring the T315I mutation, and in adult patients with CP-CML resistant to and/or intolerant of first-and second-generation TKIs.
