CARsgen Announces Data of Allogeneic BCMA CAR-T Cell Therapy CT0596 for Relapsed/Refractory Multiple Myeloma at the 2025 ASH Annual Meeting

SHANGHAI, Dec. 8, 2025 /PRNewswire/ — CARsgen Therapeutics Holdings Limited (Stock Code: 2171.HK), a company focused on developing innovative CAR T-cell therapies, announced that clinical data from its allogeneic BCMA-targeted CAR T-cell product candidate, CT0596, for the treatment of relapsed/refractory multiple myeloma (R/R MM) was presented in a poster at the 67th American Society of Hematology (ASH) Annual Meeting. The poster was titled “A First-in-Human Study of CT0596, an Allogeneic CAR T-Cell Therapy Targeting BCMA, in Patients with Relapsed/Refractory Multiple Myeloma.” The publication number was 2296.

This clinical trial (NCT06718270) enrolled 8 patients with R/R MM in the dose-escalation phase who received CT0596 infusion. The median number of prior lines of therapy was 4.5 (range: 3-9). Enrollment was not restricted by NKG2A expression levels. Regarding lymphodepletion, 6 patients received full-dose lymphodepletion with fludarabine (30 mg/m²/day) and cyclophosphamide (500 mg/m²/day) for 3 consecutive days, while 2 patients received reduced-dose lymphodepletion. CT0596 was administered at dose levels of 1.5×10⁸ (n=1), 3×10⁸ (n=5), and 4.5×10⁸ CAR-T cells (n=2), with one patient receiving two infusions.

As of August 31, 2025, all 8 infused patients were evaluable for efficacy, with a median follow-up of 4.14 months (range: 0.9-7.9 months). Six patients achieved a partial response (PR) or better: 3 achieved complete response/stringent complete response (CR/sCR) (all in the full-dose lymphodepletion group), 1 achieved very good partial response (VGPR), and 2 achieved PR. Among the 6 patients who received full-dose lymphodepletion, 5 achieved PR or better. Six patients in the full-dose lymphodepletion group achieved minimal residual disease (MRD)-negativity at Week 4. Patient 01 maintained ongoing sCR and MRD-negativity as of Month 8. Patient 04 achieved PR with resolution of extramedullary disease following the second infusion. CAR-T cell expansion was observed in all 8 patients. Among the two patients who received the 4.5×10⁸ dose, one achieved sCR, and the other exhibited deepening response to VGPR.

CT0596 demonstrated a manageable safety profile. Four patients experienced Grade 1 cytokine release syndrome (CRS); no Grade 2 or higher CRS was observed. No immune effector cell-associated neurotoxicity syndrome (ICANS) or graft-versus-host disease (GVHD) was reported. No dose-limiting toxicities, treatment discontinuations, or deaths were observed.

The study is still in the dose-exploration phase. The lymphodepletion regimen has been determined, and higher cell doses are being explored to further define the recommended dose (RD). The company plans to initiate a Phase 1b registrational study for CT0596 in 2026.

The overall response rate was 100% (95% CI: 76.8, 100.0) with 11 (78.6%) patients achieving complete response (CR) or stringent complete response (sCR). All patients who achieved CR or better were minimal residual disease (MRD) negative at 10⁻⁵ threshold. The median progression-free survival (mPFS) and the median duration of response (mDoR) were 44.1 months and 43.2 months in CR/sCR patients, respectively. The median overall survival (mOS) was not reached. The patient survival rate at 60 months post-infusion was 76.9%.

Socceroos match times set for 2026 FIFA World Cup clashes with USA, Paraguay and European rival

The Socceroos’ World Cup adventure has been confirmed as being a west coast of North America ride, with a trip to Canada, a journey to California and a marquee match against hosts the United States in Seattle.

The dates, times and venues for Tony Popovic’s team were confirmed on Saturday in Washington DC, the day after the draw for the 2026 event, which has pitted Australia against the USA, Paraguay and the winners of a European playoff — one of Turkey, Romania, Slovakia or Kosovo.

That match against European opposition will be first up, on just the third day of the tournament in Vancouver, Canada, at 9pm on Saturday, June 13 — that’s a good early-afternoon slot for Australian TV viewers on Sunday, June 14 at 2pm (AEST).

There will be much hoopla over the match against the hosts USA in Seattle’s Lumen Field, home of the NFL’s Seahawks, at 3pm on Friday, June 19.

That means fans in Australia will have to set their alarms for a 5am AEST kick-off on Saturday, June 20, to watch the glamour tie of Group D.

The third match has been confirmed against South American side Paraguay for 7pm on Thursday, June 25, at San Francisco Bay Area Stadium in Santa Clara, California, which is a midday (AEST) kick-off on Friday, June 26.

Evening kick-offs in North America dominate the complete schedule, which was released on Saturday, with FIFA having an eye on the expected heat in the US, Mexico and Canada, while also providing most fixtures in prime European evening viewing time.

The final takes place at 3pm local time, when the heat may play a factor in the open MetLife Stadium in New Jersey on July 19.

The opener between Mexico and South Africa in the roofless Azteca Stadium, scene of the 1970 and 1986 finals, in Mexico City is also an afternoon kick-off.

Trump administration waives $11 million fine for Southwest Airlines’ 2022 holiday meltdown

The Trump administration said it will forgive the last $11 million of a civil fine against Southwest Airlines that stems from the carrier’s 2022 holiday meltdown that stranded some 2 million passengers after nearly 17,000 flights were canceled.

The Transportation Department under the Biden administration in late 2023 fined the Dallas-based airline $140 million, though the agency then credited the airline with all but $35 million of that amount because of its compensation to customers.

In an order filed Friday, the Transportation Department cited Southwest’s more than $1 billion investments in its technology and operation since the holiday meltdown as reason for the additional credit. Southwest was due to make the last payment next month.

Man’s simple Christmas gift sparks chain of eerie ‘Godwinks’ — then a final twist stuns everyone

SQuire Rushnell and Louise DuArt, co-founders of the “Godwinks” brands based in Massachusetts, shared those thoughts with Fox News Digital as well as the following true story.

On a cool morning some years ago just a few Sundays before Christmas, Chris Wright was driving his family to church in Athens, Georgia, when he noticed a woman walking alone on the side of the highway, carrying a gas can.

That image haunted him. After dropping off his wife and kids, he turned around to see if the woman needed help.

Spotting her still walking on the berm, he pulled his car over, lowered the window and asked the woman if she needed assistance. 

The look on her face shifted from fear and worry to surprise and relief.

As he drove her to the gas station, she told him she was so thankful. She was going through a bad patch, she said, with only $5 in her purse. She was worried about buying Christmas presents for her child and putting food on the table.

Wright filled her gas can and drove the woman, named TunDe Hector, back to her car. 

As he was about to leave, he decided to give her what he had left in his wallet. Handing her $40, he apologized that it couldn’t be more. 

“Be nice to someone today.”

She teared up and thanked him. She also silently thanked the Lord for answering her prayers. 

She said to Wright, “Can I hug you?”

As he drove away, he felt a surge of gratefulness — that he had even a little something to help someone in need so close to the holidays. He also recalled that his mother had always told him, whenever she sent him off to school in the morning, “Be nice to someone today.”

When illness hits

Three years after that incident, Wright’s mother was diagnosed with a serious illness

The hospital said she could go home if she had a nurse’s aide to help her. The aide originally scheduled for the job couldn’t make it, so a substitute arrived.

The substitute aide’s name was TunDe Hector.

From the very beginning, Wright’s mother was drawn to Hector. Her warmth, kindness and gentle care brought comfort during a very difficult time.

When Wright stopped by one day, his mother introduced him to Hector. She said she’d worked as a nurse’s aide on and off for several years but had always dreamed of becoming a nurse.

Then she asked Wright if he attended church. When he said yes and named the church, her face lit up.

“I met a young man from that church,” she said excitedly. 

She talked about the time she’d run out of gas weeks before Christmas, and was walking along the highway when a man pulled up, said he was taking his family to church when he saw her and circled back. He filled her gas can and gave her money when she really needed it.

Wright gasped and said, “That was me.”

The words flashed through his mind before he said them aloud: “Wow, what are the odds?”

Again, just like before, they hugged.

Beyond their dreams

Over the next few weeks, as Wright’s mother’s health declined, Hector was her rock. The two women formed an unbreakable bond.

When his mother passed away — on TunDe Hector’s birthday, of all days — Wright and his family wanted to honor the caregiver’s devotion. 

They started a GoFundMe campaign, hoping to raise $1,000, to help Hector pursue nursing school. But her story touched so many that the campaign grew beyond their dreams.

When Wright handed her a check for $35,000, Hector nearly collapsed in tears, overwhelmed by the love and generosity surrounding her.

Today, Hector has completed her nursing school training. She learned that her employer is going to pay 100% of her tuition for law school. 

Her plan is to specialize in advocating for patients and home health care.

Cities hit hardest by crime, poverty rank among America’s least relaxed, study finds

With many Americans leaving high-cost, high-crime cities behind, a new study found that daily life in some U.S. communities is significantly more “relaxed.”

A new ranking by LawnStarter shows that many pockets of calm exist far from urban centers, which are ripe with stress and instability.

The study analyzed 500 cities on 42 metrics related to well-being, including sleep quality, mental health, commute times, access to nature, financial stability and recreation. 

Wealthy suburbs and tech hubs with high incomes and reliable local amenities help shield residents from some of the stresses many other parts of the country deal with, the lawn care service’s report found.

San Jose took the top spot among large cities, mainly for its mental and physical well-being scores. San Francisco and Seattle, which have ample outdoor access and high salaries, followed closely behind. Suburban havens in midsize cities also topped the list, including Sunnyvale, California; Naperville, Illinois; and Carmel, Indiana.

Smaller cities such as Newton, Massachusetts; Mountain View, California; and Woodbury, Minnesota, also ranked highly with strong scores in mental health, finances and social support.

LawnStarter noted that 26 of the top 30 cities have median household incomes of $106,000 or more, well above the national median of about $84,000.

At the bottom of the rankings were cities such as Flint, Michigan; Dayton, Ohio; and Detroit. They had high smoking rates, elevated blood pressure, inadequate sleep and long-term economic strain drag down quality of life, according to the findings. Other cities faring poorly, such as Memphis and Cleveland, battle high crime, traffic fatalities and persistent poverty.

While California cities dominated the top tier thanks to high incomes and positive wellness factors, parts of the Pacific Northwest, like Spokane, Washington, and Eugene, Oregon, landed on the opposite end with some of the nation’s highest depression rates, a trend often linked to long, dark winters and seasonal affective disorder.

Many large urban centers face intense pressure from record-high housing costs, deteriorating infrastructure and illegal immigration surges that strain public services, according to multiple reports. As a result, many Americans have already fled high-cost cities in search of safer neighborhoods, more space and a lower cost of living.

The New York Post reported that both New York City and Los Angeles have seen major worker losses as residents flee to cheaper, lower-stress cities in the South.

Residents are especially fleeing California, South Florida, Long Island and New Jersey “in droves” for the South, according to a July report from personal finance magazine Kiplinger. Those moves are primarily driven by surging housing costs, overcrowding and a sharply higher cost of living. Nearly two-thirds of movers went to lower-cost Southern and Sun Belt cities, the outlet reported, places that often overlap with the nation’s most relaxed and least stressed communities.

“Public policies that provide income security, safe housing, good nutrition, health insurance and family-friendly workplaces would go a very long way in reducing stress nationwide,” Boston University sociology professor Deborah Carr said in the LawnStarter report. “However, that is a big wish list that is difficult to achieve.”
